For issues making a backup in iTunes, the take a look at the article below to walk you through troubleshooting the issue on your computer. It will have you try backing up on a different user as well as resetting the lockdown folder.

Does that ring any bells?

The only other thing I can think of is trying a different cable and USB port. If that does not work then the next best thing would be to contact Apple for further assistance.
