Since installing the new software update -Mac OS X Lion 10.7.5- I can no longer use hot corners, shortcuts and mission control. This is incredibly frustrating. Any suggestions / help would be much appreciated.

This sounds weird but if you are using a non-apple wireless mouse & dongle, try unplugging then see if hot corners work with the built-in touchpad.
Have you tried just restarting your computer? My hot corner for mission control suddenly stopped working as well. I restarted my Macbook Pro and it started working again.
