Currently I use iPhoto library manager to store all my photos into library for different years. When the new photos app replaces iPhoto will it transfer all my photos in each iPhoto library in one go, or will I have to do it several times?
Photo will not merge your iPhoto Libraries. If you want them merged then do that first with Library Manager.
Will the new version of photo be able to cope with loads of photos i.e. not go slow like iPhoto can do when it gets too large ?
Define "loads"? I have 60k in iPhoto with no problems.

iPhoto can handle up to 1000000 photos. If you are experiencing performance issues, the problem must be caused by something else. Perhaps you don't have enough free storage on your system drive? Then MacOs X will be generally slow.
1 - Photos is not released so no one knows exactly how the released version will work
2 - with iPhoto usually it is a a really bad decision to have multiple libraries - it always always is best to have one larger library
3 - 100 GB is not a large library - mine is about 4 times the size and contains about 70,000 photos
