I just updated my MacPro (Late 2013 - Dual D700s) from 10.10.1 to 10.10.3 and all of a sudden my MacPro won't recognise the USB hub on the Dell UP2414Q. Every USB device I had connected to the display was not seen by the Mac. And the integrated card reader won't work as well. This surely worked before the update as I had my mouse connected to the display.

Try resetting the monitor to factory settings via its display setting menu. I have same Mac and monitor and I just tested mine and had same problem. Resetting the monitor just resolved the problem for me.
Just tried that but resetting did not help – still no USB.
On the monitor you did go to Menu>Other Settings>Factory Reset>Reset All Settings?
Do the USB ports work on the Mac?
Try resetting the SMC and NVRAM?PRAM
