System Preferences > Startup Disk
to deselect and reselect your start up disk.

I have only a grey screen with a fille who is blinking within a question mark I can't start the system !

As cdhw tried to explain, it cannot find the startup volume. That article should help resolve the issue, but it could be happening because the hard drive is malfunctioning, corrupt, or dead.
