Q: I have an iPhone 5 S running IOS 6.0.1. If I update to IOS 8.3, will I loose anything (i.e., photos, contacts, etc) during the update? iPhone has not been synched to my Mac Book Pro. Thank you.

Yes, you might loose some files if you don't backup and sync properly. Follow these instructions carefully.

About backups in iCloud and iTunes - Apple Support

Note that your iTunes backup needs to be Encrypted, or your sensitive information will not be transferred. Also note that your photos or videos are not part of the backup, you need to Import them into your computer photo app.

Well, it is time to sync it up with the Mac.

Safest way to upgrade an iPhone is thru iTunes on a Mac. Configure iTunes to sync everything and all your stuff on the phone will be saved on the Mac (photos, contacts, etc.). Set the backup option in the Summary pane to be kept in local storage (not the iCloud), with password. Then tell iTunes to do the iOS update. At the end, when the phone does the final reboot to start using the new iOS, you will probably get an error from iTunes saying the phone did not respond and it timed out. Ignore it, but you will have to unplug and replug the cable for iTunes to see the phone again when it finally starts.

Just beware that you will need as much room on the Mac as stuff the phone has, cause you're backing the second on the first. Especially since you have not synced the two.
