If you are getting the screen that shows the USB cable and connecting to iTunes, it is too late to backup. That screen means it has entered the recovery mode which automatically erases all content.

You need to use the recovery mode instructions for connection as given in http://support.apple.com/kb/HT1212 and summarized:
