i have bought a used iPhone 5s but unable to determine if it has already screen protector in place or not. How to find if the iPhone 5s has a screen protector in place?

Look closely with a magnifier, especially around the Home button and around the edges of the screen. If there is something there, it will look like an additional layer (the screen protector) on top of the screen's glass.

if I place a finger on the camera, it feels smooth. Does the front camera without screen protector can feel the edges of the camera?
There is always (as far as I know) a hole cut for the front camera on a screen protector..otherwise, you are taking photos through a plastic sheet (the screen protector). This is a great way to check and one I didn't think of. If the screen is smooth where the front camera's lens is and you feel no cutout or ridge, I'd say there is no protector on the device.
I just felt my iPhone 5s screen (which DOES have a protector on it) and can definitely feel where the screen protector is punched to allow for the camera lens.
