Ensure that you have Wrist Detection turned on:
Apple Watch app on iPhone > My Watch > General > Wrist Detection - on
The hourly Stand goal is achieved when you've "stood up and moved around" for at least one minute during that hour.
Apple Watch uses an accelerometer to identify movement, so a minute's walk around with your arms swinging naturally will achieve this.
This needs to be repeated within each of 12 separate hours throughout the day to achieve the daily Stand goal (closing the blue Stand ring on your Activity app icon).
If you've checked out the info in Jonathan's reply and it's still not working, reset the watch (hold the crown and side button for a few seconds until you ss the Apple logo). That will often kick a glitchy feature into action - you don't lose any data
Make sure the stand reminder is "on": Apple Watch app > My Watch > Activity > Stand Reminders > "on"
The risk protection activation and hard reset seem to do the trick...thanks for your support

Except that when I do stand in response to the reminder, no matter what I am doing, including walking around, the Watch does not issue the "You did it" message until 4-5 minutes has passed.
You may also find it helpful to calibrate your Apple Watch.
Calibration helps your watch to more accurately interpret data from the accelerometer:

That happens to me sometimes - the "You did it" message might take from a few seconds to a few minutes to arrive. I'm not bothered by it, because my overall Stand performance doesn't seem to be affected. It's probably worth reporting it to Apple at http://www.apple.com/feedback if it does concern you
