How do I completely start over with a hard drive?

I have a hard drive that I need to partition to be bootable. There is currently a 1TB hard drive that I boot from. I have a new 6TB hard drive that I wish to boot from with all of my information from the 1TB hard drive. I must partition the new hard drive into GUID to be able to be booted from. How do I do so when Disk Utility won't let me? Is there a way to start completely from scratch?

Mac Pro (Mid 2012), OS X Yosemite (10.10.4)
