When i connect my iPhone 6 I am able to transfer photos from my Phone to the PhotosLibrary on the USB, but when i Sync, the photos library doesn't get highlighted. So unable to sync. more

It sounds like you are unable to sync photos located on your external drive. I would need a little more information on your configuration to provide a better answer. However, let's try to sync your photos by going through the folder section via the article below. You would just need to direct iTunes to where your photos are stored on your external drive.
If your photos are in a folder
- Select "Choose folder" from the pop-up menu.
- Choose the folder where you store your photos.

- Click Apply.
You can organize photos into subfolders to create photo albums.

We found that my Photos Library file was damaged and that also led to the issue of not able to copy or sync. We repaired the file by using Command+Option key pressed and double clicking the Photos Library file. It prompted me to start repairing.
