All of my iTunes music is on an ext. hard drive connected to my home network through a router. Is there any way that devices that are part of my iTunes home sharing network can access music on this ext. hard drive without my laptop computer being on?

Additional information: The laptop computer is what I used to setup all the music, via iTunes, that is located on the external hard drive connected through the home network router.

I don't know exactly what you mean by "devices". If you mean other computers then presumably you could set it up so their iTunes libraries used the same media files. If you mean to things like iPhones and iPads via Apple's Home Sharing then no, because your iTunes is acting as a server for that and no server, no sharing.
