Can I use Skype on my TV from my Mac computer while using Apple TV?

If you have mirroring enabled and working properly and you had your microphone and cameras positioned correctly, then yes you can do that.
I often do it with FaceTime and my iPhone, but it's possibly much easier putting the phone immediately below the TV so that everyone is looking towards the camera.
