Of course it might be something else, like some sort of technical problem with your feed, but obviously I can't diagnose that without being told its URL.

I have created a new podcast and have uploaded 2 episodes on the same day (podcast rss feed - http://simplecast.com/podcasts/1769/rss & podcast page - https://thesea-townpodcast.simplecast.fm/episodes) but when iTunes picked up the feed, only the most recent one posted (iTunes page - https://itunes.apple.com/us/podcast/the-sea-town-podcast/id1089833365?mt=2),missing one episode entirely and now they are going to be out of order.
How do I get iTunes to pick-up the other episode?
Can I only post 1 podcast per day or expect to experience this again?

Episode 2 is in the feed and appears for subscribers, so it looks as if this is another Store glitch - several people have reported slowness in updating. You should certainly be able to post more than one episode a day (though it's better not to as subscribers may miss one for automatic download). This certainly looks like a version of the same Store problem and there isn't really anything you can do at your end. You could report the problem to Support: go to https://itunespartner.apple.com/en/podcasts/overview and click the 'Contact Us' link at the bottom of the page
