Why isn't there enough space to back up my iPhone??

My PC has 74 GB free space. My iphone 6s (running iOS 9.3.1) has 64gb capacity but still has 29 gb free. iTunes says there is not enough space to back it up. What can I do? I have freed all the space I can afford to on both the PC and the iPhone.
