2. Yes, if that drive contains a partition or disk image formatted as Mac OS Extended.

And yes you can back it up by dragging it to a separate drive that is formatted Mac OS extended (journaled) - if that is your only backup you would be much better off to use TimeMachine with its default settings so you always have a current backup rather than an occasional random backup - and also have a backup of everything on your computer
