Will I lose my music/videos if I delete them from my "downloads" folder after I have transferred them to my iTunes?

The normal behavior for iTunes is to put a copy of media you add manually in the correct iTunes media folder, so in theory you can delete those downloads. If you want to double check you can right click on the item in the iTunes window and > Show in Finder to reveal the actual file iTunes is using.
