Verify your sound is not muted by adjusting the volume.

Music or other sounds from the same speakers can easily washout the mail sound.

Verify your settings in three locations:

>System Preferences>Sound

Mail>Preferences>General>mail sound

>System Preferences>Notifications>Mail>Play sound for notifications

If both of those locations are verified, toggle them off and on again, try quitting mail and restarting your machine—launch mail.

Sometime re-installing the combo update on top of your existing 10.11.5 resolves small system glitches such as yours.

Go to system preferences > notifications follow the screen shot settings .
