Thanks for visiting the Apple Support Communities. Since the App Store works on the iMac when connected to IPv4, the issue seems to be within the settings for IPv6.
The first article I can offer to assist you is the article from Apple: What is IPv6? At the end of that article, there is a link to test your IPv6 connectivity. It has you visit Test your IPv6. Also, ensure your configuration is properly set up using this: Configuring IPv6 in OS X
If you are able to connect, but the App Store is still the only connection that fails, you will need to verify the ports the App Store uses. Using this article: TCP and UDP ports used by Apple software products I see that the App Store uses port 80 and port 443.
You can test if those ports are open using this article: Testing TCP port connectivity between computers and devices.
You may also wish to review these articles if you are unable to resolve your issue. I added the AirPort articles too in case you are using an AirPort device or get one of those errors.
