How can I sync my iPhone 6S when I try and get a message that iTunes could not backup the iPhone because not enough free space is available on your computer?

Well... how much free storage space do you have on the computer?
Taken at face value, it means that you have to delete some files on your computer because there isn't enough free space on the hard drive for the backup. Your computer hard drive needs some free space for fragmentation so if you are done to a few precious GB of free space, you can't use all of what is left.
However, you haven't provided any details at all so I am just guessing.
