Any application that can poduce graphics will work with a drawing tablet:
Keynote, Pages, Pixelmator, Illustrator, Autodesk Graphic, Open office, the list is endless.

How do you draw on Keynote? It will just advanced slides.
How do you draw on Pages? It will just highlight text.
Pages has a pen tool (for drawing lines). And yes, you can use the pen of the Bamboo tablet for that. Or for any application that produces graphics – just like Gary says.

Pages pen tool is not meant for free hand drawing. It is bezier.
Bezier yes, but you still can use the pen of your tablet to set the anchor points. There's a lot of ways you can use a tablet: I use mine as a mouse replacement. 🙂
But in stead of bickering about details: why don't you just get the demo version of Pixelmator, and give it a try?

You asked for an app that can be used for drawing on a Wacom tablet. Pixelmator can do that. The principle is the same as in Photoshop.
The problem is that both apps are limited to drawing anything within only those apps. When doing a presentation, you want to highlight or circle text. Or draw on the actual presentation. Neither app has any type of overlay ability (For Macs, I don't know one that does).
I guess this is a limitation of Macs that PCs don't suffer from. With PCs, you can use a tablet/pen and draw on other stuff without any problems. Powerpoint does this very well. Keynote doesn't do it at all (there's no tablet/pen integration for this type of drawing in Keynote. No one doing presentations is going to use bezier curves).
Please start a new topic in which you make this clear in the first post. Your question was way to general to expect a useful answer. Include the words on-screen annotation.
Will do. I tried to market you last response as the answer since it does answer which apps you can draw in using a Mac. But the forums aren't marketing it. Any who, thanks.

Keynote is not a player. It is a presentation authoring application with a full set of drawing tools to create shapes, lines, text and graphs.
Pages uses the same drawing tools as Keynote.

Your now asking a different set of questions, however the application for this job is Skitch. It is specifically used in live demonstrations, it has the following tools; shapes, arrows, freehand marker, highlight, text . Look at the website here: Skitch
