If yes, it should show when it last updated (just below the description under iCloud Photo Library and just above "Optimize iPhone Storage" on that same settings screen.
"iPhoto" does not support iCloud Photo Library -- the newer "Photos" app does.
iTunes and WiFi sync are not involved in iCloud Photo Library and neither is Photo Stream.
The only thing you should need is:
1 - On the Mac: Apple menu > System Preferences > iCloud > Photos > iCloud Photo Library turned on (and optionally Optimize Mac Storage in the Mac Photos app Preferences)
2 - On the iPhone: Settings > iCloud > Photos > iCloud Photo Library turned on (and optionally Optimize iPhone Storage)
and 3 - a wi-fi connection.

iCloud Photo Library is available with iOS 8.3 and later, Yosemite 10.10.3 and later, and tvOS 9.2. You can also access your entire photo library from the Photos app on iCloud.com.
