If I understand your issue, you have never been able to back up your iPhone 6 Plus using iTunes for Windows. Since backups are extremely important, I would suggest you first back up your iPhone using iCloud, and then continue troubleshooting your iTunes for Windows issues.
How to back up your iPhone, iPad, and iPod touch
iCloud
- Connect your device to a Wi-Fi network.
- Tap Settings, then scroll down and tap iCloud.

Scroll down, tap Backup, and make sure that iCloud Backup is turned on.

- Tap Back Up Now. Stay connected to your Wi-Fi network until the process completes.
- Make sure that the backup finished: Tap Settings > iCloud > Storage > Manage Storage, then select your device. The backup should appear in the details with the time and backup size.
Get help making a backup or manage your available space in iCloud.
If your iTunes backup couldn‘t be completed or you can‘t restore from a backup
Take care,

At this point, I think it would be helpful for one of our technical advisors to work with you one-to-one to help you get your iPhone backed up using iTunes for Windows. Or, if you live near an Apple Retail Store or an Apple Authorized Services provider, and advisor can help you backup your iPhone to iCloud in person. Click here to initiate that support.
Cheers,
