I certainly understand how important it is to be getting and sending mail. Especially getting it going after an update. There's a few things we can check out.
With the steps you may have tried, have you seen this article: If you can’t send or receive email on your Mac - Apple Support
If you've done most of the steps, double check the last one to run the Mail Connection Doctor:
3. Check your connection in Mail Connection Doctor. It might be able to tell you more about the issue.
Let us know any results and we can from there.

Many thanks for the swift reply. As it turns out, the problem is caused by my router, not the Mac (apparently, it only happened coincidentally at the time of my OS upgrade). I connected my MacBook Air (still on 10.11.3) and got the same problem. For some strange reason, the router only acts up with the Macs, but not the iOS devices. If I use different means of internet connection (i.e. my 4G iPad as Wifi hotspot), all works fine. I have contacted my internet provider, who has provided the router (with integrated cable modem).
