Thank you for using the Apple Support Communities!
It is my understanding you are not able to use your built in camera. I have a couple suggestions that hopefully will help you.
The first thing I would do is backup everything that is on the computer. Nothing I am going to suggest should cause any data loss, but it is always a good idea to have a backup. Your computer has software on it called Time Machine. It is fantastic software for backing up your computer. Information on how to use Time Machine is available here:

It is important to try to isolate what is causing this to happen. The first thing to do to help isolate what is causing the problem is set up a test user account and see if the problems still occur. Follow all the instructions that you find on this page:

If the issue does not happen in the test user account, then you are going to want to check the login items in the account that is having the issue. Try removing the login items and see if the camera starts working.

As suggested i did try creating new user account & also removed login items. Problem still persists.
The camera button & the effects button in photo booth ..both are not usable
is there something else which needs to be tried.

Since the camera is not working in either of the user accounts and does not work for any programs, it is looking like it may be a hardware issue.
I would suggest having it looked at either at an Apple Retail Store or an Authorized Apple Service Provider. You can find the nearest one here:
Be sure you back the computer up before taking it in for service.
