If that hasn't helped set it back to where it was and try once more. Otherwise see the second box in Troubleshooting issues with iTunes for Windows updates and see if a full tear down and rebuild of iTunes helps. Note the recommendations to disable antivirus when reinstalling iTunes and to run the installer as the administrator.
If you still have issues exclude the iTunes folder from any real-time anti virus scanning. If you don't have a backup of your iTunes library and other important data make them now just in case the skipping is an indication of underlying problems with the drive.
