Sep 15, 2016 10:45 AM in response to techbot93by x64uk,I'm seeing a similar thing. I was attempting to re-address poor playback quality (which I have done in the past by changing to Directsound) and found the settings locked.
I first noticed the issue after the 12.5.1 update, but cannot rule out the issue was introduced with a Windows 10 upgrade to Redstone 1 (the "Anniversary" update) which I performed about a week earlier. (Windows 10 x64 v 1607).
Are you using the Anniversary update? Did your preferences work between the Windows 10 upgrade and the itunes update?

The steps in the second box are a guide to removing everything related to iTunes and then rebuilding it which is often a good starting point unless the symptoms indicate a more specific approach. When reinstalling right-click the downloaded installer and use run as administrator.
Review the other boxes and the list of support documents further down the page in case one of them applies.
The further information area has direct links to the current and recent builds in case you have problems downloading, need to revert to an older version or want to try the iTunes for Windows (64-bit - for older video cards) release as a workaround for installation or performance issues, or compatibility with third party software.
Your library and device backups should be unaffected by these steps but there are links to backup and recovery advice should it be needed.

Everything was fine until the other day, when I updated iTunes. Initially I had issues with iTunes "not seeing" my iPhone 6 so had to reinstall after troubleshooting for a while.

Does the behavior persist in iTunes' own safe mode, or if you right-click the iTunes icon and use run as administrator, or in a different user profile?

(Oh some extra info in response to tt2's suggestions - "Run as administrator" and "logged on as different user account" - no improvement. iTunes safe mode - no improvement.)

Sep 15, 2016 12:34 PM in response to turingtest2by techbot93,I held in ctrl+shift and all I got was a UAC notification, to which I gave it permission. No notification of safe mode. That didn't work. New user account, didn't work. Another clean uninstall using IObit uninstaller and file deletion. Fresh install, no luck.

What AV are you running? Any change if you temporarily disable it?
Can you disable UAC temporarily in case that is the problem?
Are you on a corporate network with group policy object lockdowns?

Sep 15, 2016 1:29 PM in response to turingtest2by techbot93,No, just a home installation of Windows 10 Pro. I'm running McAfee Total Protection. I already disabled "real time scanning" to try. And have added the iTunes executables to the "whitelist". I ran the program as admin on tests earlier so I doubt the UAC is to blame. Thanks for your help though. It will just be a borked line of code I bet, I don't really want to try a fresh install of Windows 10 to be honest over something so small. It wasn't a case of me updating from Windows 7 either. Everything has been fine for the most part since the clean install last year. I've done SFC too

Sep 15, 2016 2:06 PM in response to techbot93by turingtest2,Use the iTunes Feedback form and include a link to this thread so they can see what troublehooting steps you've already taken, or register with developer.apple.com and make use of Apple Bug Reporter to report the issue. They don't generally talk back, but they do claim to read everything.
