Thanks for visiting Apple Support Communities.
It looks like music videos don't play after updating iTunes to the latest version. I'm sure you want to get back to watching videos, so let's see what we can do to solve this.
Since this was working before, we can create a new user account on your Mac and test the behavior there. You can find a guide to follow at How to test an issue in another user account on your Mac.
When it comes to AirPlay, you may want to take a look at "Requirements for using AirPlay" in Use AirPlay to display video from your Mac on an HDTV.
Let us know if the issue persists in a new user, as well as more information about the videos you're playing and the Mac you're using, and we'll go from there.

Sep 26, 2016 2:14 PM in response to sarah_s1by awyg18,I have this problem too - or at least the bit about iTunes 12.5.1.21 videos not playing, and particularly, video playlists not working.
I have music videos in a playlist under 'Movies'. Since this and the previous version of iTunes, the play button for the overall playlist no longer shows. And if I click the Play icon at the top left of the screen nothing happens. But if I double click one of the movies it does play... but the remaining videos in the playlist don't play.
I have OSX 10.11.6 on a 2012 mac mini, 16GB ram.
