From the Mail menu bar click Mail > Preferences then select the General tab.

Then make a selection from the: New Messages Sound pop up menu.

And open System Preferences > Sound then select the Sound Effects tab.

Make sure: Play user interface sound effects is selected.

Thanks for your reply, but I have tried your suggestion and it still does not work.  I still can not add any new sounds as I did in El Captain. New Messages is just what it says and you can not add any new sounds

In typical Apple fashion. When they add and also take away. There used to be an option of add custom sounds to Apple mail alerts.  For example,  a custom .wav file is played when  an email is received.  That option is gone in Sierra. Really Apple Engineers?

You are absolutely correct, but I found a way. Go to the library in the Go menu. Then select the sound folder and add the the WAV file and reboot. The new sounds will now appear in the e-mail sounds folder. Then select the sound and you are set to go.

Accept starting in El Capitan Apple started using SIP to protect system folders , so SIP needs to be temporarily disabled to allow copying files to the "Sounds" folder.  Directions are here : How to modify System Integrity Protection in El Capitan | Macworld

You do not need to disable SIP to copy into your home directory, only if you you are trying to alter anything within the System section, it's to protect system files after all.

The proposal that skpnsf has posted has always been the way I have added sound files to my apps, but it needs to be the Sounds folder within you Home directory and not under Macintosh HD/System/Library plus it needs to be a compatible file (I use wav) - you also should not need to reboot. Just open Mail after moving the file into ~/Library/Sounds (or if you do not have a Sounds folder just create one) and it should appear at the bottom of the "New  message sound" section within mail Preferences.

I've just tested this works on my iMac so hopefully you also should be able to use the same method.

That's a good suggestion, unfortunately it doesn't work for me. The only way I can get a custom sound it by placing it in the System/Library/Sounds folder.  Not sure why the option to pick a custom sound inside of Mail was removed in Sierra.
