It is recommended by Apple that when Apple Watch is used in or around water that the user activates water lock mode as this prevents accidental water damage to your Watch.
So, in direct answer to your question, Yes activate water lock before you Use Apple Watch in the shower

Whilst showering with Apple Watch Series 2 is OK, Apple recommends that the watch should not be exposed to soap (or to shampoo, conditioner, lotion or perfume), as this can negatively affect the watch's water seals and acoustic membranes.
The watch should also be cleaned with fresh water and dried with a nonabrasive, lint free-cloth after coming into contact with anything other than fresh water.

No - it is not necessary to enable Water Lock before showering whilst wearing Apple Watch Series 2.
If preferred, enabling Water Lock beforehand locks the screen, which may prevent accidental interactions.
Otherwise, to manually clear water from the speaker after the watch gets wet under other circumstances than tracking a swimming workout:
Swipe up on the bottom of the watch face to open Control Center > tap on Water Lock > turn the Digital Crown to immediately unlock the screen again and clear any water from inside the speaker.
