It may help to ensure that you have stopped directions, when you have finished following them: open the Maps app on your watch > press firmly on the display > tap on Stop Directions.

I had a friend with this problem for 2 days and when the last update came in it fixed it self.
Update the software on your Apple Watch - Apple Support
Here is a link to help you go through the updating process.
My friend also had problems syncing with his car until the update as well. Did you happen to run into that problem?

@Jonathan UK: unable ti check tonight, but, i remember that when the GPS route / track is finished, you don't have the possibility to press and select "stop directions"
You only have the foutr items:
search
nearby
contacts
transport
(or something like that, i am in a french version)
So ti be able to use locate me again, you have to start a new GPS route / track (or something like that, i am in a french version), then press firmly on the display > tap on Stop Directions.
Only after that you are able to use it again.

tthe issue doesn't appear when you stay on the indications but only if you show the map on the watch.
indications on the screen-> no issue
map on the screen -> issue !
Probably a bug in watchOS 3.0
