If I’m understanding your post right, you have been unable to take screenshots on your iPhone 6 after updating to iOS 10.0.2. I take screenshots on my iPhone all the time, so I can certainly understand reaching out for help when it's no longer working for you. I'm happy to help you with this.
First, make sure you are following the steps in How to take a screenshot on your iPhone, iPad, and iPod touch, which explains that you must press and hold the Sleep/Wake button first and then press and release the Home button. If you're doing it the other way around, or pressing the Sleep/Wake button and the Home button simultaneously at the exact same time, it would not work for you. Also, check the storage on your iPhone as well. If you don't have enough storage to save the screenshot, it would not allow you to take one. You can check to see your available storage in Settings > General > Storage & iCloud Usage.
If you have ample space on your iPhone and you're attempting to take a screenshot using the correct steps, but the issue still persists, the next step would be to power your phone off completely, and then power it back on. Try taking a screenshot once your phone is powered on again.
