Can i use my iphone during backing up?

If you mean during an automated iCloud backup, no. The phone must be locked.

If you mean during a manual iCloud backup, I would say this... even if you can, don't. To interrupt a data backup is risky for possible data corruption and just the amount of time it would likely add to the overall process.
