Even with the headphones on, my phone is still on loudspeaker mode. It seems that the phone does not detect the headphones. What to do? I've cleaned the jack and it still not working. The headphones is working at other phones, I've also tried using other headphones but still not working, always on loudspeaker.

Yes you can use the lightning headphones on your iPhone 6s Plus, as long as you have iOS 10. Follow the steps below.
If your headphones aren't detected
If you plug in your headphones, but sound plays from your device's speakers, try plugging in a different pair of headphones. If the same thing happens, get your device inspected at an Apple Retail Store or Apple Authorized Service Provider, or contact Apple Support.

You should be able to use the iPhone 7 headphones (or the adaptor) on your 6s Plus although I have not tried it. Your headphone jack is broken. And before you break the Lightning port or adapter, do not wrap the headphones around your iPhone with the jack still plugged in.
