I keep my Iphone back-up thorough Itunes on my Mac Book. But the backup is on 45 GB which takes up most of my comp. hard drive. What can I do to compress it? Or can I move the back up location to my wireless, external hard drive, so when I back up my phone on mt computer at home it places the file on the external one?

You cannot compress the backup. You can reduce the size by copying off data that gets backed up and then removing it from the device, e.g. the contents of the camera roll. See About backups for iOS devices - Apple Support for more.
To move the backup data to a different drive see Relocate iOS device backups. The process might only work for a locally connected drive.
