I am using Windows 10 and have Itunes set as default player. Is it possible to preview or in this case prelisten to an audio file when selecting it in windows explorer, when itunes is set as default music player?

Not with iTunes. iTunes will only play content added to the library. Windows doesn't have an independent preview player. You could install MediaMonkey as an alternative player for this task and access it from the context menu while leaving iTunes as the default player.
