I'm just trying out the Audirvana Plus trial version, but I am unable to hear a difference between the same record 16/41 and 24/48 directly in Audirvana-iTunes in my Mac late 2009 through a pair of Yamaha MSP5A Monitor speakers. Is an external DAC required to listen the difference?

Are you sure the FLAC you started with was 24-bit (sometimes labeled as FLAC-HD)?
If it was a normal CD-quality (16-bit) FLAC, inflating it will not improve the audio quality.
Yes, I am sure because is the same track downdloaded from Society of Sound (Bowers & Wilkins) in both resolutions. Apple Lossless 16/44.100 and FLAC 27/48.000. Both also appear clearly in the Audirvana´s window.
On second thought, if you are playing through Audirvana, the iTunes settings don't really matter. 😟
