Hi, I have a new Samsung Portable SSD T5 connecting via USB
After a drive test, I am getting speeds of 420mbps from my Macbook Air
However, when I test it on my Mac Pro Tower (Mid 2010), the speeds are more like 40mbps
Does anyone know why this might be? I'm using the exact same cable

Ah, yes I just checked, the Tower is USB 2.0.
I'm using a USB3 to USB-C cable
Would it help if I got a USB 2 to USB-C cable?

No, you can't make the computer-end run any faster with different adapters.
It would help the speed in the Mac Pro tower if you got a USB-3 card to fit in a PCI-e slot.
The built-in USB ports on the Mac Pro tower are only USB 2 while I suspect ( you did not give moidel date) the MacBook Air's USB ports are USB 3
