No.
USB 3 came out in 2012 iMacs. As for Thunderbolt 2, I'm not sure.
No USB 3 and Thunderbolt 2 are not upgradeable, those ports are on the main logic board.
Thunderbolt 2 was Late 2014.
Thunderbolt 3 was 2017
