This icon has appeared and is not going away even when I close all tabs. The Mute button does not seem to do anything and does not respond with an action. How can I remove this???

Since the screenshot that you provided indicates the WhatsApp website is currently using your microphone, I'd like to have you quit Safari and restart your Mac:
If the microphone still appears after, visit the WhatsApp website again in Safari. Once you have the website pulled up, follow the steps under the "Customize the active website" section to turn off microphone access:
