Is this function only available when the computer is on and has been left on or should I be able to access it even after shutting down my laptop? I'm new to this and was a little unsure.

The computer needs to be powered ON so that it is able to not only ask for Touch ID but also to validate your fingerprint.
Your Touch ID button also acts as your power ON button when fully depressed.

The Mac must be powered on. Pushing (not just touching) the Touch ID pad powers on your Mac.
