How do I lock my iPad in order to do a backup?

I need to do a back up on my iPad. The directions say to be plugged into a power source, be connected to my WiFi and to have the iPad “locked”. How do I “lock” my device. Is that the same as turning it all the way off.

Locked means having the screen turned off, like it normally is when you're not using it. You can lock the screen either by pressing the power button while the screen is on, or waiting until the screen turns off. If you want to force a backup manually right now, you can do it in Settings > your name on the top > iCloud > iCloud Backup > Back Up Now. What you described are the conditions for the daily automatic backup, that will typically happen during the night.
