If you are backing up to iCloud via the Backup Now, the screen doesn't need to be locked. If you want it to have it automatically backup while charging and connected to wi-fi, you can lock the screen via the right side button -- or simply let it lock itself based on the time chosen in Settings > Display & Brightness > Auot-Lock.
