That particular setting in Do Not Disturb does no do anything unless Do Not Disturb is turned on. If Do Not Disturb is not turned on, that won't matter. Are these calls from people that are not in your contact list? If so, go to Settings>Phone>Silence Unknown Callers, and turn that off.
