Does the iPhone SE perform an iCloud backup without being connected to a computer if the iCloud Backup is turned "ON" in the iCloud settings on the iPhone SE?

Yes. iCloud Backup does not use a connection to a computer. See instructions in the iCloud section of: How to back up your iPhone, iPad, and iPod touch - Apple Support
