When I'm on a call, the screen locks and I cannot unlock it no matter what buttons I press. How do I stop the screen from locking during a call?

You have a proximity sensor issue, get it checked.

No, the phone doesn't 'unlock' when I move it away from my ear. It remains disabled when I move it away from my ear to press the buttons
That is not locking, it only disables the touch using the Proximity Sensor. As soon as you move the iPhine further away from your ear the iPhone enables touch.
