Applecare Enterprise Support (Singapore) recommend a tutorial video to prepare an iPad in Apple Configurator 2.
I wonder what to fill in the 'Host name or URL', as attached below.
My school don't have any third-party program. We only have Apple School Manager and Apple Configurator 2.

That's where you enter the url of your MDM service; Whomever you choose should be able to provide that information to you.
You'll need an MDM solution like jamf or similar. My understanding is that school manager and configurator just automate device enrollment. School manager also allows you to purchase apps in bulk, etc.

Or is it possible to use only Apple School Manager as an MDM?
You can skip that portion if you don't have an MDM (though an MDM is highly recommended). If you do not use an MDM, you will only be able to manage the devices when they are physically connected to the computer you're using.
