How to set up and out of office auto message ?

Are you referring to set this up in the Mail app that comes with your iPhone? You should have told us which iPhone and iOS versions that you are using for a better answer.

This will only work if the email service that you are using, provides the Out of Office or Auto-Reply feature.

Open Settings ➔ Mail ➔ Accounts ➔ <Tap on the email account> : Here, scroll all the way down. If your email service provides the Auto-Reply feature then the option will appear here. Set it to ON. If the option does not appear then your email service does not provide this feature.
