You can use Dvorak and Colemak with a hardware keyboard without any third party add-ons. But I think they may only be available with the keyboard set to English.

Hello ~You can change keyboard language but to the options you are asking about you will need to download from App Store. Take a look here for changes you can make without app:

I think you may need to switch to English (US) to have Colemak.
To ask Apple to add this to English (UK), you can use

Tried changing the Language & Region to US english but it didn't work. iPad picks up the keyboard as UK english (which it is) and doesn't list down Colemak.

That's not the right place. Try going to settings > general > keyboard > keyboards and replace English (UK) by English (US).
Hello again~ On my iPad 8th gen. When set to English you can choose QWERTY,AZERTY and QWERTZ but that is not with hardware keyboard. Which keyboard are you using?
