Since the update to OS 11.6 the screenshot app will not capture with the first click. It is necessary to re-select screenshot and do it a second time.

I experienced this issue too.
cmd-shift-4 or 5 with Preview selected:
I suspected that macOS 11.6 had elevated the security privileges needed for Screen Recording so I tried this and it worked.
Fix:
I am not familiar with the "show floating thumbnail"
I do not have the timer set.
I have the app in my doc, and since I'm using the mouse when I want the screenshot, I do not have to go to the keyboard.
I am 99% of the time looking to capture only a "selected portion" of the screen (as per the prompt).
This whole issue has been only occurring since the update to 11.6 last week.

This looked promising, but for me it is not working. Still Preview opens, but not the screenshot. And it does not go to the clipboard either, unless I specifically also hold the control key (which then does what it is supposed to, put it in the clipboard).
Not a big deal for me, as I have Preview open all the time; but it would be nice if it had worked.
I would think that the complete solution would also pass by the System Preferences->Security & Privacy->Privacy, but the Automation tab; alas we can't add things manually there.

I am waiting to see if anyone can confirm whether this has been fixed in Monterey.
By the way, nobody should upgrade without a solid backup in place (preferably, two).
I intend to do so in a few days, but cannot afford the downtime at the moment (and also will wait to see if there is any major showstopper).
Good points, but can you please clarify where exactly in System Preferences->Security & Privacy->Privacy you added these applications?
To solve the problem where you can save to the Desktop but not to an external drive should be easy: add the application(s) to
System Preferences->Security & Privacy->Privacy->Full Disk Access

I just tested and I see the exact same thing.
I guess I would never have found it, because I use Preview constantly, and it is basically always running.
I try not to clutter up the memory by quitting all the apps that I am not currently using. Helps things to run without any fears of memory slow-down!

Screenshots here not really working since last update. Sometimes working, then not, ........... this is just poor software, thanks Apple!
Hmm...I updated to 11.6.1 and when I try to take a screen shot using shift-command-5 I get a bright blank window overlaying my screen
I have used this before as you say and know about capture/record but I hit it and got the blank light box overview
Hmm...shouldnt a screen shot be easy without all this rigamarole? And now it is not a screenshot but a screen recording? Apple should fix this!
Thanks but Command-Shift-5 used to offer a range of options with that command and it is not functional...the options you show are ok but not as flexible. Hope this returns.
Is´nt there a chance finding a reason by checking log files? Or just waiting for Monterey realease, and hope it will work then again fine?
Just in the moment my SSs did not work at all, I quit safari, mail, messages, istat menus, copyless, ...... nothing happened, but then, with some delay (so not straight after quiting one app) ...... SSs came back to work.
Then I started all these apps again, ................ and SSs are still working. This is the strange thing, if it is another app causing problems, it should quit when I start this app, .... but that is not happening.
